Description
  • The on-line coupling of LC and ICP-MS was used for fractionation and detection of species of Cu, Fe, I, Se and Zn in human serum. It has been shown that anion exchange chromatography proved better separation capability (both intra- and inter- element) than size exclusion chromatography. The mobile phases for ion exchange chromatography consisted of Tris-HNO3 buffer and ammonium salt (nitrate, acetate or formate). Formate was found to be the best mobile phase counter ion, which enables good chromatographic separation and is acceptable for mass spectrometer too. The quantitative evaluation of elements' concentrations adherent to individual fractions was performed by peak's area normalisation method. The repeatability of results ranges from 3 to 15 % (depending on the element fraction level) and represents the main part of result uncertainty. Accuracy of Cu and Zn fractions determination was confirmed by comparison with isotope dilution technique.

  • Frakcionace specií Cu, Fe, I, Se a Zn v lidském séru byla provedena pomocí on-line spojení kapalinové chromatografie a pomocí hmotnostní spekrometrie s indukčně vázaným plazmatem. Iontově výměnná chromatografie se ukázala jako metoda vhodnější než gelová chromatografie. Jako nejvhodnější mobilní fáze byl použit mravenčanový pufr. Kvantitativní vyhodnocení obsahu kovů v jednotlivých bylo provedeno pomocí normalizace píků, opakovatelnost byla 3 - 15 %. Správnost stanovení Cu a Zn byla ověřena pomocí izotopového zřeďování. (cs)
