Description
| - Novel modular methodologies for synthesis of new C-nucleosides will be developed based on cross-coupling reactions of haloaryl C-nucleosides and heterocyclications of alkynylsugars. The methodologies will be used in synthesis of large series of new C-nucleosides for biological activity screening and for applications chemical biology. Selected C-nucleosides will be incorporated to oligonucleotides and stability of DNA duplexes will be studied. Other selected examples will be converted to nucleoside triphosphates and used for studying of selectivity of incorporation by DNA polymerases. (en)
- Budou vyvíjeny nové modulární metodiky pro syntézu nových typů C-nukleosidů založené na cross-coupling reakcích halogenaryl C-nukleosidů a na heterocyklizacích alkynylcukrů. Metodiky budou využity pro syntézy velkých serií nových C-nukleosidů, u kterých bude studována biologická aktivita a které budou aplokovány v chemické biologii. Vybrané C-nukleosidy budou inkorporovány do oligonukleotidů a studována stabilita duplexů DNA. Od vybraných příkladů budou připraveny nukleosid trifosfáty a použity pro studium selektivity inkorporace DNA polymerasami.
