Description
| - The project is focused on the investigation of cryptic and pseudocryptic species diversity and differentiation of desmids (Desmidiales, Viridiplantae) based on species from model genera Micrasterias, Euastrum and Xanthidium. Strains obtained from culture collections and the isolates from different wetland habitats will be investigated. The molecular taxonomic part will be based on analyses of the 18S rDNA and ITS sequences. For molecular species delimitation, the complementary base changes in the ITS2 region acquired from its secondary structure will be used. The phenotype analyses will be based on morphological study (SEM) and the geometric morphometrics aimed at the quantitative evaluation of shape. Mating compatibility of populations will be investigated to assess data on the reproductive isolation. Using flow cytometry, we will determine the DNA content of strains. The results will be applicable for diversity assessment of desmids as the important group of freshwater phytobenthos and for evaluation of geographic distribution and ecological differentiation of their microspecies.
