  • Listeria monocytogenes je patogenní bakterie způsobující u lidí onemocnění listeriosa (úmrtnost okolo 30 %). V posledních dvaceti letech byl zaznamenán prudký nárůst tohoto onemocnění. Klasické mikrobiologické metody pro specifickou detekci L.monocytogenes jsou pracné a zdlouhavé, a dosud užívané imunochemické metody nejsou dostatečně specifické (detekují také nepatogenní kmeny Listeria sp.). Cílem tohoto projektu je vyvinout jednoduché, rychlé, dostatečně specifické, citlivé a levnéimunotechniky (enzymová imunoanalýza na mikrodestičkách, dipstick formát, imunosensor) k detekci L. monocytogenes v potravinách. Jako cílené epitopy pro přípravu specifických protilátek budou vybrány charakteristické antigenní struktury z povrchu L.monocytogenes (internalin B, peptidové sekvence z InlA, InlB a ActA). Vyvinuté imunochemické metody budou ověřeny standardními mikrobiologickými postupy. (cs)
  • Listeria monocytogenes is a pathogenic species causing listeriosis at people (the mortality rate is approximately 30 %), increasing incidence of which has been reported in the last two decades. Microbiological methods for specific detection of L.monocytogenes are time-consuming and till now used immunochemical methods are specific not enough (other, non-pathogenic, strains of Listeria spp. are also detected). The aim of this project is to develop simple, rapid, enough specific, sensitive andcheap immune-techniques (enzyme-immunoassay on micro plate, dipstick format, immunosensor) for detection of L. monocytogenes in foods. The characteristic antigenic structures from surface of L. monocytogenes will be chosen as target epitopes to producespecific antibodies (internalin B, peptide sequences from InlA, InlB, and ActA). The developed immunochemical methods will be verified by standard microbiological procedures. (en)
