| - Tato práce byla zaměřena na zhodnocení vlivu střevních bakterií na vznik zánětu. Vývoj střevního zánětu byl studován v chronickém modelu, kdy po přenosu slezinných T buněk z BALB/c myší do konvenčních SCID myší, dochází ke vzniku spontánního střevního zánětu. Bezmikrobní SCID myši byly monokolonizovány Enterococcus faecalis, Segmented fi lamentous bacteia (SFB), Fusobacterium mortiferum, Bacteroides distasonis a kombinací Fusobacterium mortiferum a SFB nebo Bacteroides distasonis a SFB. SCID myši byly také kolonizovány definovaným koktejlem (SPF). Po 8-12 týdnech byla kolitida nalezena u konvenčních SCID myší a u myší, které byly kolonizovány bakteriálním koktejlem SPF a SFB. Změny střevní bariéry v terminálním ileu byly detekovány imunohistochemicky pomocí protilátky ZO-1 (cs)
- The study was focused on the evaluation of intestinal microbiota on the development of intestinal inflammation in mice model. We used the model of chronic inflammation that develops spontaneously in the colon of conventional severe combined immunodeficiency (SCID) mice restored with the CD45 RBhigh subset of CD4+ T cells isolated from the spleen of immunocompetent BALB/c mice. Germ-free mice or mice monoassociated with Enterococcus faecalis, SFB (segmented filamentous bacteria), Fusobacterium mortiferum, Bacteroides distasonis and combination Fusobacterium mortiferum and SFB or Bacteroides distasonis a SFB were used as recipients. Mice were evaluated 8-12 weeks after cell transfer and severe colitis was present in conventional animals and in mice colonized with a cocktail of SPF microflora with SFB. Altered intestinal barrier in the terminal ileum of mice with severe colitis was documented by immunohistology using antibodies to ZO-1
