  • Studie popisuje mechanismus, jakým ATP-dependentní mitochondriální Lon proteasa zahajuje proteolýzu svých substrátů. K popisu byly použity dva substráty se známou strukturou: alfa podjednotka mitochondriální procesující peptidasy a steroidogenní akutní regulační protein. Byly identifikovány postupně vznikající peptidy a mapovány vzhledem k primární, sekundární a terciární struktuře substrátu.Bylo zjištěno, že proteolýza je zahajována mezi hydrofobními aminokyselinami umístěnými ve vysoce nabitém okolí poblíž povrchu složeného proteinu (cs)

We show that mitochondrial Lon also degrades folded proteins and initiates substrate cleavage non-processively. Two mitochondrial substrates with known or homology-derived three-dimensional structures were used: the mitochondrial processing peptidase alpha-subunit (MPPalpha) and the steroidogenic acute regulatory protein (StAR). Peptides generated during a time course of Lon-mediated proteolysis were identified and mapped within the primary, secondary, and tertiary structure of the substrate
