Description
  • Attachment of natural macromolecules to synthetic ones may lead to new %22smart%22 materials changing their properties in response to external stimuli. Synthesis and study of block copolymers consisting of hydrophilic synthetic polymers and peptides prepared by solid phase synthesis is suggested. The peptide block, thanks to its tailor-made primary structure, would enable the whole system to react to changes of its environment (e.g., temperature, pH, ionic strength). The peptide sequences designed for the synthesis of the copolymers will be derived from important natural proteins (elastin, kinesin) with corresponding structural motifs (?-helix, ß-spiral, coiled-coil). The polymer block can be represented, e.g., by poly(ethylene glycol) and the peptide part of the copolymer, e.g. by peptide (VPGXG)n , derived from elastin. Intermolecular structures (micelles, liposomes) will be formed as a result of peptide conformational changes upon increase of temperature. These properties could be utilized for preparation of new systems for controlled drug release and delivery. (en)
  • Cíle projektu jsou návrh, syntéza a charakterizace hybridních kopolymerů složených z hydrofilních syntetických polymerů a peptidů. Peptidové části budou při změně teploty či pH podléhat změnám konformace vedoucím ke vzniku nadmolekulárních útvarů studovaných fyzikálně-chemickými metodami (NMR, LS).
