"The mouse first molar (M1) is the most common developmental model in odontogenetic studies. The third molar (M3) is one of the most commonly missing tooth type in humans as well as in mice. Moreover, M1 starts its development prenatally, whereas M3 develops postnatally, therefore under different mineralization stage of the surrounding bone tissue. From this point of view, the mouse M3 may resemble the secondary human dentition. The aim of this study was to evaluate the M3 development with special focus on initial stages, particularly its origin. Methods of molar explants cultures, histochemistry and 3D reconstructions were performed to achieve the goals. The M3 tooth germ was shown to extend out from the epithelium of the second molar (M2). Moreover, the M3 encapsulation into the mineralized bone proceeds at earlier stages than M1, thus making the M3 tooth growth more difficult.
