In our laboratory, we use the maize (Zea mays) beta-D-glucosidase Zm-p60.1 that is involved in regulation of many important processes in plant growth and development. These processes can be changed slightly with mutated forms of Zm-p60.1. Protein engineering offers a means to create variants of this enzyme. These variants have changed substrate specificity towards either natural (trans-zeatin-O-beta-D-glucopyranoside, cis-zeatin-O-beta-D-glucopyranoside) or artificial substrates (p-nitrophenyl-O-beta-D-glucopyranoside, 4-methylumbelliferyl O-beta-Dglucopyranoside).
