D\u00E1le jsme studovali vliv 0,1 uM a 1 uM koncentrace doxorubicinu na sign\u00E1l p\u0159i rychlosti polarizace 1280 mV/s. DNA izolovan\u00E1 jak z rezistentn\u00EDch tak i senzitivn\u00EDch bun\u011Bk byla tedy m\u011B\u0159ena p\u0159i r\u016Fzn\u00FDch rychlostech polarizace. Z\u00EDskali jsme line\u00E1rn\u00ED z\u00E1vislosti, ze kter\u00FDch jsme ur\u010Dili sm\u011Brnice p\u0159\u00EDmek, kter\u00E9 jsme porovn\u00E1vali se sm\u011Brnicemi kontroln\u00EDho vzorku (bu\u0148ky kultivovan\u00E9 bez p\u0159\u00EDtomnosti doxorubicinu). "For monitoring the interaction of anticancer drug doxorubicin with DNA, we used the electrochemical technique of cyclic volumetry. Was measured by DNA isolated from cells sensitive and resistant to doxorubicin. We obtained voltammograms characteristic of DNA isolated from the cells. On the record, we observed two peaks. The first peak (1) is reductionist cytosine and adenine signal (potential around -1.5 V), the second peak (2) corresponds to the guanine oxidation signal (potential around -0.2 V). We also studied the effect of 0.1 uM and 1 uM concentrations of doxorubicin to the signal polarization at the speed of 1280 mV / s. DNA isolated from both resistant and sensitive cells was then measured at different speeds polarization. We obtained a linear dependence, from which we determined the lines of which we compared with the directives of the control sample (cells cultured without the presence of doxorubicin). Fig. 1 and 2 shows the average value of directives based on the concentration of doxorubicin-resistant, or sensitive cells. The vertical axis values are plotted directives (k), the columns correspond to the given concentrations. The first column corresponds to a control sample (without doxorubicin). For others, the concentration difference is not statistically significant directives.
