"The identification and quantification of plant hormones are important because of more extensive studies and usage of phytohormones in plant physiology, biotechnologists and agriculture. The method for estimation of 37 naturally occurring cytokinins is based on an immunoaffinity purification (IAC) and a baseline chromatographic separation of 20 cytokinins (trans/cis-zeatin, dihydrozeatin, isopentenyladenine, benzyladenine, meta/ortho-topoline groups) on reversed-phase column and analysis by high-performance liquid chromatography-mass spectrometry (HPLC/MS). Analyses of O-glucoside and ribonucleotide fractions were also performed by the IAC-HPLC/MS method but run separately from the basic cytokinin metabolites after enzymatic cleavage. Combination of HPLC with quadrupole-time of flight mass spectrometry (Q-TOF) was used for accurate and sensitive identification of cytokinins.
