"Some species of the genus Clostridium produce butanol during the acetone-butanol-ethanol (ABE) fermentation. To transition from acidogenic to solventogenic phase of clostridial metabolism is necessary several specific factors that influence the physiological state of the production microorganism. We chose fluorescent labelling with flow cytometric detection to determine the physiological changes of C. beijerinckii. We developed methods of fluorescent labelling with hexidium iodide, carboxyfluorescein diacetate and sytox green, that contribute to better understanding of physiological changes during ABE fermentation of C. beijerinckii. Flow cytometric detection of stained cells with selected fluorescent probes was proved to be a valuable, rapid and non-exacting method. Fluorescent detection of transition from acidogenic to solventogenic metabolic phase together with detection of amount of vegetative and sporulating cells contribute to better understanding and faster detection of acid crash, that sometimes stops the fermentation."@en .
