"A novel process strategy for the production of recombinant protein under control of AOX1 promotor of Pichia pastoris yeast has been developed during the close university-industry cooperation of partners from the Czech Republic and Switzerland in the OPTYTECH (E!3415) project. In contrast to strategies, most commonly reported in current literature (biomass growth on glycerol followed by a production phase on pure methanol) a mixture of methanol and glucose was applied in carbon-limited mode during the production phase. Since the maximum specific growth rate of Pichia pastoris on mixed substrate is superior to this achieved with pure methanol, carbon-limited addition of mixture of methanol and glucose during production phase leads to an enhancement of rate of product formation and therefore significant shortening of process duration due to higher productivities.
