"Various transition metal ions (e.g. Cu2+, Ni2+, Zn2+ etc.) bind to the imidazole side chains of surface exposed histidines of proteins. This coordination interaction (M2+ - His) has been used for protein purification by immobilized metal affinity chromatography (IMAC). If the interaction M2+ - His proceeds on the surface of an enzyme close to the active site, it can lead to efficient inhibition of the enzyme. In this study the influence of Cu2+ and Ni2+ ions on the activity of papain from Carica papayawas determined. Iminodiacetic acid (ida) and nitrilotriacetic acid (nta) were chosen as the ligands to chelate the metal ions; ida (nta) has strong affinity for these ions that ensures that the complex will not be demetalated even at high protein concen trations. It was found that complex Cu2+- IDA/NTA is specificly bound to His 159 of papain active site and inhibits papain to 50-60 % of residual activity.
