"Glass-ceramics scaffold for substitution of bone tissue was prepared from Bioglass(R) (patented by professor Hench) by thermal treatment at 1100oC for 5 hours (replica method). Glass-ceramics scaffold contains major crystalline phase (combeite - Na2O.2CaO.3SiO2) and residual glass phase (about 23%). Crystalline phase does not contain phosphorus and residual glass does not contain calcium, which is an advantage in glass-ceramics dissolution studying by in vitro testing. The scaffold was exposed to Simulated Body Fluid (SBF), which simulates an inorganic part of blood plasma and to DMEM (Dulbeco's Modified Eagle's Medium) solution, which contains both inorganic and organic part of blood plasma. Since DMEM is highly susceptible to bacterial contamination, the experiments had to be carried out under sterile conditions. Results from our previous work [1] showed that crystalline form of hydroxyapatite (HAp) was formed only in the solutions buffered by TRIS (tris-(hydroxymethyl-aminomethane)). Here we show that in the solutions without TRIS buffer (SBF 27 and DMEM) minerals such as halite (NaCl) and calcite (CaCO3) crystallizes, however, only amorphous phase containing Ca and P (ACP) is formed.
