"The principle of biochemical detection of imaginal diapauze of insect pests is aplication of tetrazolium salt solution upon the flight muscles of adult insects. The preparation is kept for one hour in darkness and after that changes in coloration visually or photometrically checked. When testing the method in the field, flight muscles are simply submerged by 0.1 mol/l solution of tetrazolium salt and kept for approx. one hour in darkness. After that the change in coloration is visually checked. During quantitative laboratory assessment the flight muscles are dissected, homogenized and incubated for one hour in the tetrazolium salt solution.
