"Biochemical assessment of acetylcholinesterase (AChE) activity is a standard protocol in biochemistry, pharmacology, and toxicology. In 1960s, Ellman\u00B4s method was developed for the purpose of AChE activity assay and it became routine after some optimizations. The method has however some disadvantages and limitations in its performance. Firstly, the used reagents are labile and fresh reagents have to be prepared at least every day. Secondly, some compounds interfere in the assay. Glutathione, thiol containing compuonds and oxime containing drugs are the most relevant. We infer to introduce indoxylacetate based assay for assessment of AChE activity. The assay was tested and basic parameters were established for the indoxylacetate. Michaelis constant and maximum reaction velocity were calculated for electric eel AChE. The assay was also performed for estimation of oxime rectivators obidoxime, pralidoxime, and HI-6 efficacy after paraoxon ethyl inhibition.
