V r\u00E1mci ozdravovac\u00EDch terapi\u00ED bylo po prvn\u00EDm kole testov\u00E1n\u00ED celkem z\u00EDsk\u00E1no 43 ozdraven\u00FDch meriklon\u016F dvou odr\u016Fd jablon\u011B (\u00B4Idared\u00B4, \u00B4\u0160ampion\u00B4), 46 ozdraven\u00FDch meriklon\u016F dvou odr\u016Fd hru\u0161n\u011B (\u00B4Lucasova\u00B4, \u00B4Max Red Bartlett\u00B4) a 9 meriklon\u016F dvou odr\u016Fd t\u0159e\u0161n\u011B (\u00B4Kare\u0161ova\u00B4, \u00B4Rivan\u00B4). Nejv\u00EDce ozdraven\u00FDch meriklon\u016F (62) bylo z\u00EDsk\u00E1no pomoc\u00ED in vitro chemoterapie. Nejm\u00E9n\u011B (13) pak pomoc\u00ED in vivo termoterapie zako\u0159en\u011Bl\u00FDch rostlin v kontejnerech. Po retestu rostlin jev\u00EDc\u00EDch se jako ozdraven\u00E9 po prvn\u00EDm kole testov\u00E1n\u00ED bylo celkem 38 % meriklon\u016F j\u00E1drovin a 67 % meriklon\u016F t\u0159e\u0161n\u011B pozitivn\u00EDch na viry, kter\u00E9 byly p\u0159\u00EDtomny i ve v\u00FDchoz\u00EDch rostlin\u00E1ch p\u0159ed zah\u00E1jen\u00EDm ozdravov\u00E1n\u00ED. "The aim of this work was to eliminate viruses from the basic plant material of fruit woody species for the system of certification and long term observation of the health status. The elimination of viruses was carried out by in vitro thermotherapy and chemotherapy. Economically important apple, pear and sweet cherry cultivars commercially grown in the Czech Republic were selected for sanitation. The presence of viruses and phytoplasmas in initial trees of selected cultivars was detected by ELISA and PCR testing before the beginning of thermotherapy and chemotherapy. Totally, we obtained 43 virus-free mericlones of two apple cultivars (\u00B4Idared\u00B4, \u00B4\u0160ampion\u00B4), 46 virus-free mericlones of two pear cultivars (\u00B4Lucasova\u00B4, \u00B4Max Red Bartlett\u00B4) and 9 virus-free mericlones of two sweet cherry cultivars (\u00B4Kare\u0161ova\u00B4, \u00B4Rivan\u00B4) in the framework of sanitation therapies after the first round of testing. The most virus-free mericlones (62) were obtained by in vitro chemotherapy. On the contrary, the least amount (13) was obtained by in vivo thermotherapy of rooted plants in containers. However, totally 38 % of pome fruit and 67 % of sweet cherry seemingly virus-free clones after the first round of testing were positive in retests for the same viruses as in the initial plants. In the case of this material, the treatment probably depresses the amount of viral particles temporarily below the threshold level of detection of used diagnostic methods. In the course of further cultivation, the virus recovered-up and re-accumulated to a level of detection. Obtained virus-free plants will be included in the established system of certification of initial plant material after repeated testing and long term observation."@en .
