"5"^^ . "25310" . . . "9"^^ . "Patients who suffered from hematooncological disease are nowadays commonly treated with autologous stem cell transplantation with high-dose chemotherapy. Severe aplastic anemia and phenomenon of spontaneous remission of hematooncological disease were observed in some patients as side effect of the treatment. After closer examination high titre of specific antibodies against ? carbonic anhydrase isoenzyme I (anti-CA I) was found in patient sera. It is expected that these autoantibodies have a protective effect on the next development of hematooncological disease. There is still unknown mechanism of their action but it is assumed to have possible inhbitory effect on the activity of carbonic anhydrase isoenzyme I (CA I). Because of that it was necessary to quantify CA I enzyme activity. For this purpose, rapid fluorimetric method for determination of CA I activity and method for determination of esterase activity of CA I were introduced. Then inhibitory effect using model system represented by enzyme and specific inhibitor (acetazolamide) was determined by measurement of esterase activity of CA I. The resulting inhibition was expressed by IC50 factor. Testing and comparison of system enzyme ? specific antibody will follow."@en .
