To understand the mechanism of toxicity in nephrotoxic compounds, a variety of in vivo and in vitro models have been used. Recently, a number of cell and tissue cultures have been developed due to necessity to limit the experiments in animals regarding economical and ethical aspects. These in vitro models have been used widely in studies focused on toxicology, physiology and biology. Most of nephrotoxic compounds causes the damage in proximal tubular cells predominantly thus the research has been focused mainly on development of cultures of these cells. The freshly isolated renal cells have been considered to be elemental in study of acute nephrotoxic effects while primary cultures have been used to estimate the toxicity for longer time period. However, the life span of primary cultures is limited and that is why the cell lines having rather unrestricted life span have been developed.
