"Basidiomycetous lipophilic yeast M. pachydermatis (1) was studied by means of indirect immunofluorescence with the aim to identify microtubular system. In contrast to other yeasts (2) it was necessary, due to a thick compact cell wall, to apply lytical enzymes, detergents and fluorescent antibodies at higher concentrations and for a longer time. The visualization of microtubules was complicated by small size of the yeast and a mutual agglutination of the cells. The presence of the cytoplasmatic microtubules in cells was successfully documented and the changes of the microtubular system of M. pachydermatis during interphase of the cell cycle were described. These changes were compared to the morphology and localization of the nucleus (stained by DAPI) and the morphology of the cells in phase-contrast microscope.The course of a nuclear division and the related role of an intranuclear mitotic spindle in this process is not known yet.
