"Many stressors influence cultivation of plants in vitro before cryopreservation and during regeneration steps after cryopreservation. Character of tissue cultures differs from in situ conditions in controlled cultivation during their growth. Today there is published a plenty of cultivation protocols for in vitro cultures of many important agricultural crops. Pre-cultivation of explants intended for cryopreservation is essential for induction of physiological state, which enables plants to withstand stressors occurring during the cryopreservation. Influences of stressors (osmotic stress, stress from dehydration) during cooling to the low temperature are necessary to be minimized for assuring of possibly best regeneration of the explants, which were cooled to the low and ultralow temperatures. Causes of damage by low and ultralow temperatures are further reviewed. Three cryopreservation methods differing in ways of the protection against abiotic stressors are mentioned."@en .
