"The primary aim of the Project is development of methods for detection and quantification of natural, accidental or deliberate contamination of food, water and environment with pathogenic microorganisms. Early detection and identification of pathogens can definitely decrease the risk of infection in humans and animals (farm animals, professional dogs, free living animals etc.). Current molecular-biology methods based on direct detection of bacterial and viral pathogens using nucleic acid amplification are the effective tool for achieving the above specified aim. Partial aims of the Project: (1) Preparation of reference material \u2013 gathering bacterial and viral isolates from various sources in the Collection of Animal Pathogenic Microorganisms (CAMP) at VRI in Brno, their multiplication and storage. (2) Development and optimization of methods for isolation of nucleic acids (DNA and RNA) from different types of matrices. (3) Development and validation of real time PCR systems for detection and quantification of the investigated pathogenic bacteria and viruses using quantitative real time PCR methods and in the case of RNA viruses, by reverse transcription and real time RT-PCR for respective pathogens and matrices. (4) Collection of field samples. (5) Verification of the developed methods in field samples. (6) Testing of the developed methods under field conditions.
