Prophylactic therapy to the central nervous system is effective in reducing the occurrence of brain metastasis in leukemias and small cell lung cancer. We hypothesized that the expression of certain genes in primary NSCLC tumors could predict brain metastasis. The expression of candidate genes will be measured by real-time quantitative reverse transcriptase polymerase chain reaction (RQ-PCR) and in given cases by cDNA microarray in 142 frozen NSCLC samples. Univariate and multivariate Cox regression analysis will be used to analyze the correlation between gene expression and the occurrence of brain metastasis. Immunohisotchemistry using tissue microarrays on independent samples will verify the findings. The expression levels of certain genes and proteins in primary NSCLC is expected to identify patients at high risk for brain metastasis, who may benefit from profylactic therapy."@en .
