Postnatal switch of glycolytic to oxidative metabolism is of crucial importance for all mammalian neonates and is essential for successful adaptation to extrauterine life. The aim of the project is to elucidate the mechanism and functional consequences of OXPHOS disturbances in premature neonates with special attention to maturation of respiratory chain complexes and their degradation. We will quantify mtDNA content in placental blood from neonates (various gestational age) and analyse respiratory chain complexes in muscle and liver tissue obtained at autopsy from miscarriages and neonates who died in neonatal period. We will analyse in tissues the expression of nuclear-encoded gene COX4 and mtDNA-encoded gene MTCO2, marker of biosynthesis, and mRNA level and protein amount of proteases calpain-2 (CAPN 2) and calpain-3 (CAPN3) induced during infection."@en .
