"The influence of candidate genes for schizophrenia (COMT, BDNF,RGS4,DTNBP,NRG1,DRD2, DRD3, 5-HT2A, SERT, GRM3, FoxP2, LMO4,MAG, G72) on the phenotypic variability will be evaluated in patients with schizophrenia. The clinical characteristics include the symptoms description, cognitive functioning, course and reactivity to antipsychotics. The neurobiological measures include the regional brain metabolism measured (18FDG PET) and structural brain parameters (magnetic resonance). To manipulate expression level of positional candidate genes with unclear function (RGS4, FoxP2, G72), in vitro technique (tissue culture) of siRNA interference will be used. The biological impact of gene silencing will be evaluated using expression profiling. As a result, we will propose the model of the clinical and biological subtypes of schizophrenia based on detected dysfunction of specific candidate genes (and their haplotypes) including adjusted treatment guidelines."@en . .
