"Glutamatergic theory of schizophrenia will be tested by the parallel animal (rat) and human (healthy volunteers) model through the application of subanestetic doses of non-competitive NMDA antagonist ketamine. We will evaluate the influence on behavior (psychopathology), cortical disconnection by the use of EEG coherence and sensomotor gating by prepulse inhibition of startle reflex (PPI), and on the expression of c-fos in rats. Model verification will be performed by intergroup comparison. Psychopathology parameters, qEEG coherence and PPI in healthy volunteers after ketamine will be compared with the same measures in schizophrenic patients. In humans, the gene polymorphisms involved in glutamatergic neurotransmission (neuregulin-1, G72, RGS4, mGluR3) will be evaluated.
