"We will study iron transport using labeling of intestinal cells by 59 Fe followed by native electrophoresis of cell lysates and detection by phosphorimaging. We will describe the intracellular iron kinetics and iron transport proteins. Transport proteins will be purified and microsequenced. Partial amino acid sequences will be used for the identification of the protein by database searching and/or for the cDNA cloning. We will construct the intestinal cell line expressing different levels of the HFE protein under the control of tetracycline sensitive promotor. These cells will be used for the studies of iron transport influenced by different levels of HFE expression. We will study the difference in composition of HFE-associated protein complexes in cells obtained from hereditary hemochromatosis patient compared to healthy cells."@en . .
