A large number of enzymes required for different modifications have now been identified in many model organisms, but despite some progress little is known about modification enzymes in Trypanosoma brucei, an evolutionarily divergent and medically very important protist. Some tRNA modification enzymes target the anticodon loop of various tRNAs and can impact cytoplasmic and mitochondrial protein synthesis. Importantly, several of them either need iron-sulfur (Fe-S) cluster or use components of the Fe-S cluster assembly pathway for their activity. The proposed aim of this project is to study tRNA modifications such as thiolation, methylation and wybutosine formation in T. brucei. Our objective is to determine their subcellular localization and their biological roles in the given compartment.
