Cathepsins that digest host blood proteins in the parasite gut represent a promising intervention target for the treatment of schistosomiasis. The main goal of the project is the characterization of schistosomal cathepsins and the development of their specific inhibitors as potential chemotherapeutics. The specific aims of the project are as follows: (a) Evaluation of cathepsins as effective targets using RNAi and chemical genomics. (b) Recombinant expression of cathepsins and their biochemical and structural characterization. (c) Rational design of selective inhibitors of cathepsins, and (d) Screening for their potency against the parasite in vitro and in vivo. This interdisciplinary research integrates the areas of structural and functional biochemistry and functional genomics and parasitology.
