"2015-02-16+01:00"^^ . "7"^^ . . . . . "7"^^ . . "LH11046" . "Abnormalities in immunoglobulin A1 (IgA1) O-linked glycosylation belong to the initial cause of IgA nephropathy. Several enzymes-glycosyltransferases (GTs) namely ST6GalNAcII, C1GalT1, C1GalT2, and GalNAcT2 contribute in IgA1 O-glycosylation which takes place in Golgi complex in a stepwise, spatially well arranged process. Aberrantly O-glycosylated IgA1-producing B lymphoblasts have decreased C1GalT1 and increased ST6GalNAcII levels. We hypothesize that abnormalities in GTs concentration and spatial distribution contribute significantly to abnormal IgA1 O-glycosylation. The first aim of the project is to localize GTs involved in IgA1 O-glycosylation within Golgi complex in B lymphoblasts from IgA nephropathy patients and healthy control to identify any abnormalities in distribution. In the second aim we propose to localize such GTs after downregulation of their expression level by siRNA to identify causative relation between abnormal GTs concentration and their localization."@en . . .
