"The cold adapted Arthrobacter sp. C2-2 employes enzyme beta-galactosidase with its activity optimum at lower temperatures compared to mesophillic organisms. So far unique 3D structure of this enzyme determined in the applicant's laboratory will be used as a starting point of further structural studies. Experiments of x-ray structural analysis will be carried out in order to determine 3D structures of complexes of the enzyme with substrate analogues or with substrate itself and with inhibitors. Acquired structural information will be utilised in comparisons with enzymes from mesophillic organisms with potential application of results to directed mutagenesis affecting the temperature profile of the enzyme and its stability. X-ray structural analysis will be used to elucidate structure of an isoenzyme from the same organism.
