"At the top of colonic crypt epithelial cells are shed into the intestinal lumen and die by detachment-induced apoptosis (anoikis), which is associated with loss of the cell-ECM and cell-cell interactions. These processes are linked with cell differentiation. However, transformed epithelial cells are resistant to anoikis to promote cancer invasion and metastasis. Therefore, we will study anoikis and its mechanisms during sodium butyrate-induced in vitro differentiation of normal and cancer colon cells. Endogenous regulators from TNF family, which mediate apoptosis, can be presented in the colon in elevated level during cancer or chronic infections. Thus, we will investigate their ability to affect signaling pathways leading to anoikis. Arachidonic acid metabolites play a significant role in the cell cytokinetics.
