"The aim is to develop an assay for testing inhibitors of dimerization of capsid proteins (CA). CA, as a part of Gag polyproteins plays a crucial role in assembly of innature capsids mediating the Gag-Gag interactions. CA dimerisation is essential also for formation of core during virus maturation. Both processes are necessary for production of infectious virions. Therefore, inhibitors of CA dimerisation would represent a novel calss of antiretroviral therapeutics which is desirable for complementation of drugs of choice for triple therapy currently used in the teatment of HIV-1 infection. We propose to clone gene encoding HIV-1 CA as a fusion protein to i) six-histidine anchor and ii) reporter protein (RP) (GFP, luciferase, etc). All constructs will be expressed and purified. An assay for quantitative measuring of inhibition of CA dimerization will be developed. This method is based on evaluation of capability of selected peptides to prefent binding of RP-CA to immobilized His-CA."@en . . . .
