"Extracellular enzymes, mostly of microbial origin, aply an important role in carbon and nutrient cycling in aquatic ecosystems. Although their function and significance have been studied extensivaly, no direct evidence of their localisation within natural freshwater seston is available. We propose to study their localisation and cell-specific activity using recently introducet fluorogenic substrates based on the fluorochrom ELF97, which precipitates at the site of enzyme activity. On the basis of our extensive previous research of exstracellular enzymes and a preliminary study with WELF97, we plan to study (1) seasonal expression of extracellular phosphatases on the taxonomic level, mainly in algae but also in bacteria and other producers of both alkaline and acid phosphatases in different water bodies, (2) expression of N-acetylhexosaminidases on chitinolytic bacteria and diatoms in freshwaters, and (3) localisation of N-acetylhexosaminidases involved in protistan grazing on bacteria."@en . . .
