We have characterized five major peptidases responsible for the digestion of host blood. These enzymes have a great potential as 'concealed' antigens for rational vaccine design. In order to evaluate the candidates to be targeted in the early phase of tick-host interaction, a dynamic expression profile of the peptidases will be determined using qRT-PCR and the activity assay. The roles of individual peptidases as well as their interrelations in the proteolytic cascade will be analyzed by functional proteomics and dsRNA interference. The ability of spirochetes to persist and multiply in these RNAi silenced ticks will be studied. The feeding success of I. ricinus nymphs on rabbits immunized with recombinant peptidases (and their combinations) as antigens will be tested with an ultimate goal to develop an efficient %22anti-tick%22 vaccine."@en .
