"Crosslinking of the high affinity receptor for IgE (FceRI) on the surface of mast cells and basophils initiates a series of biochemical events culminating in degranulation and release of mediators of allergic reactions. Early events include tyrosine phosphorylation of numerous signaling molecules, including kinases of the Src and Syk/Zap families, and rapid (within 1 min) changes in cell morphology. The link between aggregation of FceRI and changes in cell morphology remains enigmetic. In this proposal we will use rat basophilic leukemia (RBL) cells and isolated mast cells to determine physical and functional association of kinases with cytoskeletal components, mainly tubulins. Immunoblotting and immunoprecipitation techniques with monoclonal antibodies specific for the proteins under study will be used to determine kinases-cytoskeleton interactions.
