"In mammals, principal circadian pacemaker is located in the suprachiasmatic nuclei (SCN) which consist of two parts, the ventrolateral (VL-) and the dorsomedial (DM-). The molecular core clock mechanism involves interacting positive and negative transcriptional/translational feedack loops. The aim of the proposed project is to elucidate how photic stimuli entrain the molecular core clock mechanism in the rat SCN. Effect of light pulses that phase delay and advance the clock on thythmic expression of clock genes mRNA's, i.e., Per1, Per2, Cry1, Cry2, Bmall and Clock and production of their proteins as well as on dynamics of resetting these thythms in the VL- and DM- SCN separately will be studied. Expression of the clock gener mRNA's will be followed by in situ hybridization and levels of the proteins production by immunocytochemistry.
