"This project will assess the impact of sustained administration of varying doses of morphine and/or methadone on opioid and adrenergic signaling in the rat heart. A special attention will be devoted to investigating the presumed cardioprotective or adverse effects of these drugs. We shall analyze the cellular compartmentalization of myocardial G-protein-regulated signaling and the possible alterations induced by opioids in cellular/membrane distribution of opioid and adrenergic receptors, trimeric G-proteins and adenylyl cyclase, as well as functional status of the whole signaling system. The presumed changes in protein expression and/or distribution will be analyzed by a proteomic approach. This research should extend our knowledge about the molecules and molecular mechanisms ingaged in the development of cardioprotection.
