Insect make their steroid hormones ecdysones from cholesterol in a process whose regulation is poorly understood. We cloned a gene ecdysoneless, responsible for ecdysone synthesis in the fruit fly Drosophila. Using fly genetics, we will examine the function of this potentially important steroidogenic factor. During molts and metamorphosis, ecdysone induces through its nuclear receptor a cascade of transcription factors, genetically studied in Drosophila. Non-drosophilids such as moths would be important models to study ecdysone response if they could be genetically manipulated. Aiming to perturb and thus define gene functions in vivo, we transformed silkmoths with a heat-inducible expression vector. We use it to address roles of key steroid response genes by their misexpression, dominant negative forms and RNA interference, all methods that would be of versatile use."@en .
