. . . . "Persistent problem of the protein crystallography is low efficiency of the crystallization process. Polymer precipitants used in our structure determinations of 30 protein complexes showed high efficacy. They often proved to bind partly at specific sites on the protein surface and partly they remain solvated in buffer filling 40-80% of the crystal volume. Preliminary analysis shows that the efficacy of these precipitants is high and that its function consists in selective blocking the potential adhesion sites on the surface of the target macromolecule. Protein surface possess a number of potential adhesion sites, however the crystal of diffraction quality appears when only a single adhesion site is active during crystal growth. The project analyzes experimentally and theoretically this phenomenon, i.e. quality of adhesion sites, types of blocking ligands and conditions for their use.
