The goal of these studies is to develop a systematic and complex theoretical background to the synthesis of novel platinum complexes with unique DNA binding activities capable of overcoming the problem of cellular resistance to platinum drugs already used in the clinic and of their limited activity against the most common tumors. An important aspect is to examine and compare the mechanism of action of platinum complexes structurally distinct from those used in the clinic. The anticancer activity of platinum complexes arises from their ability do damage DNA. Therefore, the systematic studies will be performed aimed at analysis of DNA binding modes of a series of novel platinum complexes by biophysical and biochemical methods.
