"Palmitoylation is one of the most frequent posttranslational modifications, which has an important regulatory function in many G protein-coupled receptors, and it may presumably condition preferential partition of some receptors into membrane domains (DRMs). In the present project we will aim to determine sites of palmitoylation in delta-opioid receptor (DOR) and to evaluate the role of palmitoylation in targeting of DOR to DRMs. Different mutated DOR, in which cysteins will be individually or in combination replaced with alanines, will be stably expressed in HEK293 cells and analysed for palmitic acid incorporation and targeting to DRMs. We will also fuse DOR (and its palmitoylation-defficient mutants) with caveolin or flotillin in order to investigate the ability of these typical DRM-associated proteins to drag such chimeric DOR constructs into DRMs. The functional aspects of DOR-initiated signalling (coupling to G proteins and adenylyl cyclase) will be assessed in parallel."@en . .
