"The aim of the project is an analysis of several cysteine and aspartic lysosomal proteinases (predominantly cathepsin C a D) on different structural levels (primary structure, posttranslational modifications - especially glacosylation, tertiary structure) together with a determination of their specificity and interaction with inhibitors. Inhibitory topic is based on a characterization of novel natural protein type inhibitors. The research is focused on their spatial structure determination (by X-ray crystallography) and description of interaction mode with cognate proteinases. The project will provide fundamental information for a better understanding of lysosomal proteinases action and intracellular targeting rules, and, finally, the role of lysosomal proteinases in human pathogenesis as well as their inhibitory regulation."@en . .
