Here we propose to study the 3D structure of the scaffolding protein p12 and the 13 kDa form of M-PMV protease (PR) using isotopicaly aided NMR spectroscopy. The role of the C-terminal extension of PR on its structure and higher proteolytic activity compared to the 3D structure of 12 kDa PR will be determined. We will also investigate the role of PR truncations in the virus life and the liberation of the M-PMV PR from the polyprotein precursors and its activation using tissue culture experiments. Using bacterial and in vitro assembly system we will determine the molecular mechanism and structure/function relations of Gag-derived domains involved in the assembly of retroviral capsid."@en . . .
