The enzymatic activity of mite allergenic proteases (MiAPs) plays a critical role in the allergic reaction. This project focuses on structure-function relationship in MiAPs and regulation of their activity. The novel MiAPs will be purified and characterized by proteomic and enzymological methods, and their proteolytic interaction with immune molecules will be evaluated. The natural modulators of activity of MiAPs will be investigated, namely endogenous protease inhibitors and polysaccharides. Further, new synthetic inhibitors of MiAPs will be designed with the help of molecular modeling and combinatorial chemistry. The selectivity of the inhibitors for the target MiAPs versus relevant human proteases will be analyzed. The project will provide new information about molecular mechanisms of mite allergen regulation, and tools for the potential therapeutic application."@en .
