"This project proposes a novel and powerful approach for the efficient analysis and mapping of large eukaryotic genomes. Specifically, we propose the development of means for the construction of chromosome-specific DNA libraries, isolation of chromosome-specific DNA sequences and markers, and the development of chromosome paints for economically important cereals (Zea mays, Secale cereale, Hordeum vulgare and Triticum aestivum), legumes (Vicia faba, Pisum sativum, Medicago sativa), a coniferous tree (Picea abies), and a dioecious plant Melandrium album. The project is based on a multidisciplinary approach combining classical genetics (the use of isogenic and translocation lines) with flow cytogenetics (chromosome isolation and flow-sorting of single chromosome types), molecular techniques (DNA isolation, cloning and primed in situ DNA labelling).
