The aim of this project is to elucidate how auxin signal triggers ABP1-mediated changes at the PM and subsequent changes in vesicle trafficking. Microscopical tracking of membrane vesicles carrying fluorescently-tagged auxin transporters and other membrane proteins that undergo recycling from PM will be performed in Arabidopsis roots and various transgenic tobacco cell cultures. By this approach it will be addressed how specific the ABP1 action is. Since the signal transduction downstream of ABP1 is still poorly understood, the activity of PM-related phospholipases will be addressed as potential target of the ABP1-driven nontranscriptional auxin effects. The auxin-induced protoplast swelling assay will be used to follow ABP1-dependent responses in Arabidopsis mutants in genes coding for various phospholipases and for other candidate ABP1-interacting proteins."@en .
