"Recently, we have described and characterized functional consequences of the first human mutation in the transmembrane iron transporter DMT1 which is responsible for severe hypochromic microcytic anemia. Due to the severity of the disease it is impossible to analyze the molecular pathophysiology of this mutation in details. As an alternative we propose to study mk mice who also carry a homozygous missense mutation in DMT1. The goal of the proposed project is to characterize the consequences of DMT1 mutation on erythropoiesis. We plan: 1. Study the bone marrow erythroid progenitors using hematopoietic colony assays. 2. Analyze possible outcome of erythropoietin therapy for mk mice anemia and the dependence of this therapy on the presence of heme in the diet of mk mice. Our work will improve our understanding of normal and pathological erythropoiesis which is essential for the development of novel therapy strategies in patients with similar defects of erythropoiesis."@en .
