"Most cancer related deaths are associated with advanced disease and metastasis. Epithelial-mesenchymal transition (EMT), in which epithelial cells lose their polarity and become motile mesenchymal cells, is viewed as an essential early step facilitating dissemination of tumor cells. Recent studies are suggesting a role of MDM2 in the regulation of cell motility, invasion and metastasis. Using benign and transformed epithelial cells, the aim of this project is to elaborate on our original preliminary data and analyze the causes and consequences of the EMT-associated down-regulation of MDM2. We will analyze the role of transforming growth factor-beta signaling pathway and EMT-regulating transcription factors in the regulation of MDM2 expression as well as the impact of MDM2 on the expression of EMT regulators. The results will be supported by functional analysis of cell migration and invasion. The main output of the proposed project will contribute to the recognition of possibilities to modulate signaling pathways leading to EMT and cancer progression."@en .
